Here is the text of the Bacterial pathogenesis abstract which was presented by the Karolinska research group. Note the emphasis on TLR2 and TLR4, just like we had in our presentation.

TOLL-LIKE RECEPTOR EXPRESSION AND CYTOKINE SECRETION AFTER IN VITRO STIMULATION OF LUNG AND BLOOD CELLS IN SARCOIDOSIS

Background: Pattern-recognition receptors (PRRs), such as toll-like receptors (TLRs), induce innate immune responses to conserved structures of microbes. We asked whether differences in TLR expression or response to stimulation of TLR2, TLR4 or the intracellular PRR NOD2 are associated with sarcoidosis.


Methods: Bronchoalveolar lavage (BAL) cells were obtained from sarcoidosis patients (n=8) and blood cells were obtained from patients (n=15) and healthy controls (n=8). Cells were incubated overnight with the TLR2 ligand peptidoglycan (PGN), the TLR4 ligand lipopolysacharide (LPS) and the NOD2 ligand myramyl dipeptide (MDP) or medium alone. TLR2 and TLR4 expression on blood monocytes and BAL macrophages were measured by flow cytometry and cytokines (IL-1 , IL-6, 1L-8, IL-10, IL-12 and TNF-alpha ) were analyzed using a combination of cytometric bead array and flow cytometry.

Results: TLR2 expression on monocytes was higher in patients compared to controls. Stimulation of monocytes with MDP resulted in higher TLR2 and TLR4 expression in patients than in controls. LPS, MDP and PGN downregulated TLR2 expression on macrophages in BAL but upregulated TLR2 expression on blood monocytes. LPS downregulated TLR4 expression on macrophages to a higher degree than on monocytes. No obvious differences were seen between patients and controls regarding cytokine profiles.

Conclusions: Differences between patients and controls were observed both in baseline TLR2 expression and with regard to TLR2 and TLR4 expression following in vitro stimulation of NOD2. These findings suggest that an altered pattern-recognition receptor function may play a role in the pathogenesis in sarcoidosis.
